MTHFR activates 5,10-methylenetetrahydrofolate to 5-methyltetrahydrofolate (5-MTHF). While MTHFR is a key enzyme involved in serotonin, dopamine, and catecholamine production, catechol-O-methyltransferase (COMT) is the enzyme responsible for degradation of catecholamines.     COMT has also been demonstrated to play a role in estrogen metabolism through inactivation of the catecholestrogens. Catecholestrogens are formed during the metabolism of estrogens such as estradiol. Catecholestrogen inactivation decreases the cancer-causing potential of these metabolites, while simultaneously increasing the amount of 2-methoxyestradiol, a metabolite that has been shown to inhibit the growth of breast cancer cells.

    Identifying both MTHFR and COMT status is often helpful. People with reduced COMT activity may be more sensitive to folate supplements, as folate can increase neurotransmitter levels. If both MTHFR and COMT are slow, it’s usually best to address COMT first.
